Summary
In vitro test for the quantitative determination of the catalytic activity of AST GOT (EC 2.6.1.1; L‑aspartate: 2‑oxoglutarate aminotransferase) in human serum and plasma on COBAS INTEGRA systems. This method sheet describes the application for AST activated by pyridoxal
phosphate (test ASTPL, 0‑594). The application for AST without pyridoxal phosphate is described in the method sheet Aspartate Aminotransferase. Method according to the International Federation of Clinical Chemistry (IFCC),with pyridoxal-5′-phosphate.3,4 AST in the sample catalyzes the transfer of an amino group between L‑aspartate and 2‑oxoglutarate to form oxaloacetate and L‑glutamate. The oxaloacetate then reacts with NADH, in the presence of malate dehydrogenase (MDH), to form NAD+. Pyridoxal phosphate serves as a coenzyme in the amino transfer reaction. It ensures full enzyme activation.
